中华普通外科学文献 （ 电子版 ） 2011 年6月第 5 卷第 3 期 Chin Arch Gen Surg （Electronic Edition），June 20 11 ，Vol 5 ，No.3 ·247 · · 讲座与综述 · 神经型钙黏附蛋白与恶性肿瘤的关系 王敏 尚培中 【摘要 】 神经型钙黏 附蛋 白 （N-cadherin ） 在上皮恶性肿瘤中表达上调， 且与上皮-间质转换关系密 切 ， N-cadherin 高表达提示肿瘤 的能动性和转移能力增强 ， 在肿瘤的发生、 浸润、 转移 、 血管生成 中起 了 重要作用。ADH-1 作为 N-cadherin 抗体 的一个全身性抗癌药， 诱导靶细胞的凋亡 ， 破坏肿瘤组织 中的 血管， 通过破坏胞外 N-cadherin 间的黏附 ， 改变 N-cadherin 和成纤维细胞生长 因子受体 （FGFR-1 ） 间的 关系， 破坏 FGFR-1 ， 导致 FGFR-1 表达缺失 ， 从而破坏肿瘤血管 ， 抑制肿瘤 的生长 ， 为临床肿瘤 的治疗 提供了理论基础。 【关键词 】 神经型钙黏附蛋白； 上皮型钙黏 附蛋白； 上皮 - 间质转换； 上皮肿瘤 Relationship between N -cadherin and malignant tumor WA NG Min, SHA NG Pe i -z hong. Graduate School of He be i North College, Zhang j iakou 075000, China Corresp onding A uthor: SHA NG Pe i-z hong, Email sp z 251@163.com 【Abstract 】 The expression of N-cadherin is upregulated in epithelial malignant tumor and is associated with epithelial to mesenchymal transition.The overexpression of N-cadherin expression has been shown to promote motility and invasion. N-cadherin plays an important role in the tumorigenesis, infiltration, metastasis and angiogenesis.ADH-1 as a systemic anticancer agent can induce apoptosis of the target cells and cause tumour vascular damage,which disrupt extracellular N-cadherin adhesion, altering the association of N-cadherin and FGFR-1, and destabilized FGFR-1 to induce the expression of FGFR-1 lost,and the result was that the tu- mour vascular was damaged and the growth of tumor was inhibited.
