凋亡相关基因PDCD5表达与SLE临床相关性的研究 赵丽娜,赵杰,,张秀军摘要 目的:通过检测(programmed cell death 5PDCD5)在系统性红斑狼疮(systemic lupus erythematosusSLE)中的表达,探讨其与SLE发生与发展、自身抗体产生和肾脏损伤的关系。方法:采用RT-PCR和Western blot,分别从mRNA和蛋白两个水平检测PDCD5在67例SLE患者和50例健康对照外周血单个核细胞(PBMC)中的表达。结果:SLE活动期患者PDCD5阳性表达率和表达水平显著高于正常对照组(P0.01);活动期患者PDCD5 的表达水平显著高于稳定期患者(P0.01),阳性表达率相比无显著差异;SLE稳定期患者PDCD5 的表达量显著高于正常对照组(P0.01),但阳性表达率也无显著差异。有2种或2种以上自身抗体阳性的SLE患者PDCD5表达量显著高于有1种或无自身抗体阳性的SLE患者(P0.01);狼疮性肾炎患者和非狼疮性肾炎患者的PDCD5表达无显著差异(P0.05)。结论:SLE患者PBMC中PDCD5的表达上调,活动期患者升高更为明显,随着自身抗体种类的增多,PDCD5的表达量亦随之增多,提示PDCD5可能通过增加表达量而导致异常的淋巴细胞凋亡参与SLE发生和发展。 关键词 系统性红斑狼疮;PDCD5;凋亡;外周血单个核细胞 apoptosis-related gene PDCD5 and SLE ZHAO Li-Na, HE Bao-Ling, ZHAO Jie, ZHANG Xiu-Jun Department of Biological Sciences of North China Coal Medical College, 063000 Tangshan, China [Abstract] Objective To study the expression of programmed cell death 5 (PDCD5) gene in systemic lupus erythematosus (SLE) patients and analyze the association with the occurrence and development of SLE, autoantibody generation and kidney damage. Methods The expression of PDCD5 mRNA and protein in PBMC from 67 SLE patients and 50 healthy controls were detected by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) and Western-blot. Results The positive rate and expression lever of PDCD5 from active SLE patients were significantly higher than that in normal controls (P0.01). The expression lever of PDCD5 from active SLE patients was significantly higher than that of inactive SLE patients (P0.01). Nevertheless, No significant difference of PDCD5 positive rate was found between active SLE patients and inactive SLE patients. The expression lever of PDCD5 from inactive SLE patients was significantly higher than that of normal controls (P0.01), while there was no significant difference in PDCD5 positive rate between inactive SLE patients and normal controls.
