水蛭的化学成分研究 1 1,2 3 1 荆文光 ，符江 ，刘玉梅 ，刘安 * （1.中国中医科学院中药研究所，北京，100700；2.江西中医药大学，南昌，330004 ； 3.西南交通大学，成都，610031 ） [摘要] 目的：提取、分离、鉴定水蛭化学成分。方法：采用硅胶柱色谱法、 Sephadex LH-20 柱色谱以及制备HPLC 等方法进行分离纯化，根据波谱学数据 和理化性质进行结构鉴定。结果：从水蛭甲醇提取物中分离鉴定了18 个化合物， 分别为胆甾醇（1）、十六烷基甘油醚（2 ）、菜油甾醇（3 ）、烟酸（4 ）、尿嘧啶（5 ）、 尿苷 （6 ）、次黄嘌呤 （7 ）、次黄嘌呤核苷 （8 ）、黄嘌呤 （9 ）、苯丙氨酸 （10）、 丙氨酸 （11）、腺苷 （12 ）、脯氨酸 （13 ）、缬氨酸 （14 ）、异亮氨酸 （15 ）、丙三 醇 （16 ）、棕榈酸 （17 ）、琥珀酸 （18 ）。结论：化合物4 ~ 6, 8 ~ 10, 12 ~ 14, 16, 17 均首次从水蛭中分离得到。 [关键词] 水蛭；化学成分；生物碱；氨基酸 Research on Chemical Constituents of Hirudo nipponica Whitman 1 1,2 3 1 JING Wen-guang , FU Jiang , LIU Yu-mei , LIU An * （1.Institute of Chinese Materia Medica, China Academy of Chinese Medical Sciences, Beijing 100700,China; 2.Jiangxi University of Traditional Chinese Medicine, Nanchang 330006,China; Southwest Jiaotong University, Chengdu, 610031, China） [Abstract] Objective: To extract, separate and identify the chemical constituents from Hirudo nipponica Whitman. Method: Chemical constituents were isolated by various chromatographic methods and were identified by physicochemical characters and spectroscopic analysis or comparison with standard compounds. Result: From the MeOH extract of the Hirudo nipponica Whitman, eighteen compounds were isolated and identified as cholesterol （1）, hexadecyl ethers of glycerol （2 ）, campesterol （3 ） , nicotinic acid （4 ） , uracil （5 ） , uridine （6 ） , hypoxanthine （7 ） , inosine （8 ）, xanthine （9 ）, phenylalanine （10）, propylamine （11）, adenosine （12）, proline （13）, valine （14）, L-Isoleucine （15 ）, glycerin （16 ）, palmiticacid （17 ）, succinic acid （18 ）.
