拳参正丁醇提取物对异丙肾上腺素致大鼠心肌肥厚保护作用 　 作者：张乡城，薛金雄，杨晓光，叶春荣，汪有彪，潘吴媛，黄志华，曾靖 【摘要】 　　目的观察拳参正丁醇提取物对异丙肾上腺素所致大鼠心肌肥厚的保护作用及探索其相关作用机制。方法采用ISO 1 mg·kg-1·d-1，背部皮下注射，连续10 d，建立大鼠心肌肥厚模型。造模第2天开始给大鼠灌胃不同浓度的拳参正丁醇提取物或NS，连续14 d，末次给药后禁食12 h，称重，麻醉后断头取血，取心脏称重后计算全心重量指数及左心重量参数；采用试剂盒测定心肌组织NOS活性及MDA含量。结果ISO模型组大鼠心脏重量参数及左心室重量指数明显提高，左心室NOS含量降低，MDA 含量升高；与模型组相比，拳参正丁醇提取物治疗组左心室重量指数下降，心脏重量指数有下降趋势（无统计学意义），心肌组织NOS活性显著升高及MDA 含量降低。结论拳参正丁醇提取物对异丙肾上腺素所致大鼠心肌肥厚具有保护作用，其作用机制与清除OFR和提高NO水平有关。 【关键词】 拳参正丁醇提取物； 异丙肾上腺素； 心肌肥厚 　　Abstract：ObjectiveTo observe the protective effect of Polygonum bistorla L. n-Butyl Alcohol Extract (PBNA) on myocardial hypertrophy caused by isopropylarterenol (Iso) in rats and explore its possible mechanism. MethodsThe myocardial hypertrophy model was established through hypodermic injection for 10 consecutive days at back skin with Iso, 1 mg/（kg·d）.Once the model was formed, the rats were administrated with various concentrations of PBNA or NS in intragastric way from the second day for 14 days. 12 h after the last administration,blood and heart were taken under anesthesia, the indexes of whole heart and left ventricle were calculated and NOS activity and MDA level of myocardial tissue were determined with kits. ResultsThere was obvious difference between Iso group and Ns group. Indexes of whole heart and left ventrical in Iso group were much higher than those in control one, NOS activity was decreased while MDA concentration was increased in left ventricle. However, both indexes of whole heart and left ventricle were decreased in BAEB group, furthermore, NOS activity was strikingly enhanced and MDA concentration was reduced in PBNA group.
