bcl-6基因非编码区结构多态性及蛋白表达及移行上皮癌关系探究 　 作者：林贞花，韩龙哲，任香善，李柱虎，宋京郁 【关键词】 ,,原位癌;基因表达;泌尿系肿瘤 　摘要］ ［目的］ 分析bcl-6基因5′端非编码区-1结构多态性及其蛋白在泌尿系统移行上皮癌及远端正常移行上皮组织中的表达情况. ［方法］ 收集47例膀胱、肾脏及尿道移行上皮癌以及20例远端正常移 行上皮组织蜡块，应用PCR-SSCP分析bcl-6基因5′端非编码区E1.11，E1.12区结构多态性，并应用免疫组织化学染色方法检测bcl-6蛋白表达情况. ［结果］ bcl-6基因5′端非编码区E1.12的结构多态性多见于Ⅰ级移行上皮癌中.bcl-6蛋白在正常移行上皮组织中的表达阳性率较低，且规则地分布在表层;在移行上皮癌组织中bcl-6蛋白的表达率较高，且分布不规则. ［结论］ bcl-6基因5′端非编码区E1.12区 域的结构多态性及bcl-6蛋白的过度表达可能在移行上皮癌的分化和分级过程中起重要的作用，但两者 无相关性. ［关键词］ 原位癌;基因表达;泌尿系肿瘤 ABSTRACT:OBJECTIVE To analyze the5′non-coding exon1of bcl-6gene and its protein expression in transitional cell carcinomas（TCCs）and normal transitional epithelial cells adjacent to the TCCs.METHODS The paraffin blocks of47transitional cell carcinomas of the urinary bladder，kidney，urethra，and20normal transitional epithelial tissues adjacent to the TCCs were investigated by polymerase chain reaction-single stranded DNA conformation polymorphism（PCR-SSCP）and immunohistochemical methods.RESULTS The positive ra- tio of bcl-6conformation polymorphism at exon1.12in gradeⅠwas significantly higher than that in gradeⅠand gradeⅢof TCCs.Bcl-6protein had a low positive rate in normal transitional epithelium，and the positive cases showed regularly expression pattern in the upper layer of normal transitional epithelia，where-as，bcl-6protein was irregularly and heavily stained in the nucleus of the TCCs.CONCLUSION Bcl-6gene conformation polymorphism at exon1.12and the over-expression of bcl-6protein may play some roles in the differentiation and histopathologic grade of TCC，but the over-expression of bcl-6protein was not related with the bcl-6gene conformation polymorphism at5′non-coding exon1.Key words:carcinoma in situ;gene expression;urologic neoplasms bcl-6基因位于染色体3q27上，具有转录抑制作用.大量的研究结果表明bcl-6基因5′端非编码区突变在降低bcl-6蛋白表达过程中起重要作用.bcl-6蛋白主要表达在生发中心B细胞及其来源的肿瘤细胞中，而且对于生发中心的形成起到非常重要的作用 ［1，2］ .据报道，bcl-6蛋白也表达在正常皮肤鳞状上皮及其来源的肿瘤细胞中 ［3，4］ ，从而推断出bcl-6 蛋白亦可能在上皮组织分化过程中起非常重要的作用.2003年本实验室首次在泌尿系统肿瘤中发现了bcl-6，bcl-2蛋白的表达，同时还发现二者具有相反的表达规律 ［5］ .
