Abstract Intracytoplasmic sperm injection (ICSI)is a new method of in vitro fertilization (IVF).As one of the assisted reproductive technique(ART) ,it have been successfully used in the treatment of choice in cases of male factor infertility and in cases of failed conventional IVF cycles in human.In dairy cow and some other animals,ICSI is mainly used to produce sexing control embryos, and to research clone and transgenic etc. This article explored and investigated on applications of ICSI technique on production of sexing control embryos in dairy cow.And analysed the current situation,approach and methods of sexing control embryos’production. Studied on effects of some factors on in vitro development of ICSI zygotes,such as cryopreservation of in vitro maturation oocytes and X-sorted sperm, oocytes’ quality rating and activation method etc..Try to provide a theoretical and practical basis for sexing control embryos’ production and application in dairy cattle. This study have there parts, the results are as follows: Part one In vitro maturation of oocytes in dairy cow Test 1 In vitro maturation of oocytes with different rating Different rating of COCs were respectively maturated in vitro according to morphology. The maturing rate of GradeⅣ COCs was 17.25%, which was lower than Grade Ⅰ(61.17%), Grade Ⅱ (48.83%)and GradeⅢ(46.77%), the differences were extremely significant(P0.01). The maturing rate of Grade Ⅱand GradeⅢ were lower than Grade Ⅰ,and the differences were significant(0.01P0.05).The average maturing rate of Grade Ⅰ, Ⅱand Ⅲ was 52.38%,which is extremely significantly higher than the average maturing rate of Grade Ⅰ, Ⅱ,Ⅲand Ⅳ(40.05%),(P0.01).This results show that the maturing rate on a downward trend with the decrease in COCs’rating.
