43株副溶血性弧菌菌体脂肪酸的成分分析( 郭川1,陈银海1,林洁敏1,陈冠武2,陈婉1 汕头市疾病预防控制中心, 广东汕头 515041;2. 汕头市出入境检验检疫技术中心, 广东汕头 515031;) 摘要:目的:了解副溶血性弧菌菌体脂肪酸组成的特征,为该菌的脂肪酸鉴定与分型提供理论依据。方法:对副溶血性弧菌标准菌株VPL4-90及42株分离株(VP-01~VP-42)进行菌体脂肪酸提取及甲酯化,通过GC-MS对脂肪酸甲酯进行分离、组分鉴定,并对菌株的脂肪酸特征进行分析。结果:所有菌株的提取物气相色谱图上都有7个峰经质谱鉴定为脂肪酸甲酯,它们分别为C14:0、C15:0、C16:1、C16:0、C17:0、C18:1、C18:0脂肪酸的甲酯;其中含量最高的脂肪酸是C16:0,而C16:1/C18:0含量较低,C14:0/C15:0/C17:0含量则最低;聚类型别上的差异。对副溶血性弧菌种内的菌型不具备鉴别能力 on Cellular Fatty Acid Components of 43 Strains of Vibio parahamolylicus Guo Chuan1, Chen Yin-hai1, Lin Jie-min1, Chen Guan-wu2, Chen Wuan1 (1. Technology center of Shantou entry exitinspection and Quarantine Bureau, Shantou, Guandong 515031, China) Abstract: Objective To figure out the characteristics of cellular fatty acid from Vibio parahaemolyticus, and provide a theoretical basis for the fatty acid typing and determination of Vibio parahaemolyticus. Method Fatty acid from Vibio parahamolylicus (including standard stain VPL4-90 and 43 isolated strains VP-01~VP-42) were extracted and methyl esterificated. GC-MS was applied to separate and determine the fatty acid methyl ester (FAME). Characteristic of the cellular fatty acid from the strains was subjected to analysis. Result 7 peaks of GC were identified by MS as FAMEs, which were the methyl ester of C14:0, C15:0, C16:1, C16:0, C17:0, C18:1 and C18:0; In the 7 components of cellular fatty acids, C16:0 was the component with highest content, while the C16:1/C18:0 were the components with lower content, and the C14:0/C15:0/C17:0 were the component with lowest content; Result of Cluster Analysis showed that the strains could be divided into 3 groups, while the Squared Euclidean distance≤9. However, the grouping could not reflect the difference of the O-serotypes.
