药新药与临床药理圆园14 年9 月第25 卷第5 期 ·595 · 质量分析研究· 基于COI 和16SrRNA 基因的地龙药材及其混淆品的DNA 条形码鉴定 马 梅，李 薇，龚 玲，卢瑞珊（广州中医药大学中药学院，广东 广州 510006） 摘要：目的 利用线 体细胞色素C 氧化酶亚基 1（COI ）和 16S 核糖体RNA（16SrRNA ）对地龙药材及常见混伪 品进行分子鉴定，探讨快速、准确鉴定地龙药材及其混伪品基原物种的方法。方法 收集地龙药材的药典收载 品种及其易混品种 10 种，提取DNA ，得到其COI 和 16SrRNA 序列。所得序列经DNAStar 校正后，用MEGA6. 0 自带的Clustal W 多重比对，除去冗余位点，比对结果经MEGA6.0 分析，构建地龙药材及其混淆品的邻接 （N-J ）树。结果 地龙药材的正品来源参环毛蚓（Pheretima asp ergillum ）、通俗环毛蚓（P. v ulgaris ）、威廉环毛 蚓（P. guillelmi ）及栉盲环毛蚓（P. p ectinifera ）与其混淆品种间COI 和16SrRNA基因序列均存在较多变异位点。由 所构建的N-J 系统聚类树图显示所测物种的单系性，即16SrRNA、COI 可以很好的将地龙药材区别于其他混伪 品。结论 所获得的COI 和16SrRNA序列可作为不同状态的地龙类药材物种鉴定的分子依据，并为进一步建 立地龙等动物类中药物种真实性鉴定体系奠定了重要的实验基础。 关键词：地龙；线 体细胞色素C 氧化酶亚基 1；16S 核糖体RNA ；DNA 条形码 中图分类号：R284.1 文献标志码：A 文章编号：1003-9783（20 14）05-0595-04 doi 10.3969/j .issn.1003-9783.2014.05.018 DNA Barcode Identification of Pheretima Medicinal Material and Its Adulterants Based on COI and 16SrRNA Gene Sequence MA Mei ，LI Wei ，GONG Ling ，LU Ruishan （School of Chinese Materia Medica ，Guangzhou University of Chinese Medicine ，Guangzhou 5 10006 Guangdong ，China） Abstract Objective COI and 16SrRNA genes were used for the molecular identification of Pheretima medicinal material and its commonly-seen adulterants ，thus to build a rapid ，accurate method for the identification of their species.
