靶向Livin基因的microRNA干扰对人卵巢癌顺铂耐药 细胞的体外影响 马彩婷1 张辉2 薛新建3 (河北医科大学 硕士研究生1 张辉：河北医科大学第四医院 妇科 博士 通讯作者2河北医科大学 硕士研究生3) [摘 要] 目的 研究靶向Livin的microRNA干扰对人卵巢癌顺铂耐药细胞的体外影响，探讨Livin与SKOV3/CDDP细胞顺铂耐药性的关系。方法 利用RT-PCR技术，检测Livin基因在卵巢癌亲本和耐药细胞中的表达水平。通过micro RNA干扰沉默SKOV3/CDDP细胞中Livin基因，利用RT-PCR方法检测Livin在异性转染组、非特异性转染组和对照组细胞中的表达；采用MTT、FCM方法测定转染后后耐药细胞在顺铂作用下细胞增殖及凋亡的变化。 结果 Livin基因在耐药细胞中的表达高于亲本细胞（p0.05）；通过MTT法检测耐药细胞在浓度分别为1μg/ml、5μg/ml和10μg/ml的顺铂在作用12h,24h,48h下的，特异性转染组细胞的增殖能力均较其它两组高，（p0.05）；FCW检测显示，1μg/ml的顺铂作用24h后，特异性转染组细胞凋亡指数明显高于其它两组（p0.05）。 结论 Livin基因可能参与卵巢癌细胞顺铂耐药，靶向Livin基因microRNA干扰有可能够逆转卵巢癌顺铂耐药细胞的耐药性。 [关键词] 卵巢癌；Livin；microRNA；耐药 [中国图书馆分类法]：R711.75 ［文献标识码］ A The effect of micro RNA interference target Livin on chemosensitivity of SKOV3/CDDP cells to CDDP in vitro Ma Caiting1, Zhang Hui2, Xue Xinjian3 （Hebei medical university Master1 Hebei medical university the forth hospital Department of gynecology2 Hebei medical university Master3） [Abstract] Objective： To transfect human ovarian carcinoma drug-resistance cells with micro RNA against Livin , study the effect of down regulation of Livin on cisplatin resistance of human ovarian carcinoma drug-resistance cells.
