[Abstract] Objective: To compare the staining effect of alkaline methylene blue staining method with improved Papanicolaou
staining method on vaginal smear of SD rats cellular tissue, and to find a relatively simple and effective staining method for
identifi cation of estrous cycle. Method: Vaginal smear of 40 female SD rats were made continuous for 6 days in every morning.
Each sample was stained synchronously by the two methods respectively, and then the results were analyzed after observation.
Result: The results of the two staining methods were basically consistent. The process of alkaline methylene blue staining method
was simple and saved time and material. The cellular morphology of improved Papanicolaou staining method were clear with
obvious color separation and good transpar
