降钙素基因相关肽对兔成骨细胞增殖和TGF-β1表达的影响＊( 周 燕，张 纲，卓贤露，裘松波，谭颖徽 （重庆400037，第三军医大学新桥医院口腔科） [摘要] 目的 检测外源性降钙素基因相关肽(alcitonin gene-ralated peptide，CGRP)对体外培养兔成骨细胞转化生长因子-β1 (ransformation growth factor-β1，TGF-β1)表达的调控作用，探讨其对成骨细胞增殖的影响。方法 将新生兔头骨成骨细胞进行原代培养，用不同浓度的CGRP分别在不同时间段处理后，用MTT法检测细胞生长活力增殖情况用estern blot检测成骨细胞TGF-β1的蛋白表达。结果 成功分离并培养兔颅骨原代成骨细胞，进行碱性磷酸酶（ALP）染色鉴定成骨细胞。结果同一浓度的CGRP随着时间的增加MTT值呈增加趋势；而就同一时间点做比较，随着CGRP浓度的不断增加，MTT值也呈上升趋势，并且在48h和72h，这种趋势愈加明显（P0.05）。另外随CGRP浓度的增加TGF-β1条带灰度显著升高，显示TGF-β1蛋白的表达逐渐增强（P0.05），CGRP可能促进兔成骨细胞TGF-β1的表达。结论 CGRP可能通过调节成骨细胞TGF-β1蛋白的表达促进成骨细胞的增殖分化从而影响骨代谢。降钙素基因相关肽成骨细胞转化生长因子-β1成骨细胞细胞培养体外实验A experiment study about the effects of calcitonin gene-related peptide on cultured osteoblasts proliferation and tumor necrosis factor-β1 Zhou Yan, Zhang Gang ,Zhuo Xianlu, Qiu Songbo, Tan Yinghui （Department of Stomatology, Xinqiao Hospital, Third Military Medical University, Chongqing 400037, China） Abstract] Objective To study the possible association of extrinsic CGRP with the expression of TGF-β1 in cultured rabbit osteoblasts. Methods Rabbit osteoblasts were cultured and then different concentrations of CGRP were added, the proliferation of osteoblasts were detected by MTT ,and the expression of TGF-β1 were detected by Western blot. Results The rabbit osteoblasts were primarily cultured. And alkaline phosphatase (ALP) dyeing appraisal of osteoblasts.With the increase of the concentration of the same time MTT value was the increasing trend; And the same time point comparison, along with the increase of concentration, MTT value also shows ascendant trend, but in 48 h and 72 h, this trend more and more obvious.With the increasing of the concentration of CGRP TGF-β1 strip gray significantly increased, show TGF-β1 protein expression increased (P 0.05), CGRP may promote the rabbit osteoblasts TGF-β1 expression. Conclusion CGRP might promotes the proliferation of cultured rabbit osteoblasts by regulating the expression of TGF-β1. [Key words] calcitonin gene
