结论：EML4-ALK融合基因是非小细胞肺癌中关键而又独立的分子靶点，对其结构、临床病理联系、ALK抑制剂及其耐药机制的的进一步研究，可以为非小细胞 肺癌患者提供更多的治疗选择。【关键词】 非小细胞肺癌；EML4-ALK 融合基因；分子靶向治疗；克里唑替尼Clinical meaning of EML4-ALK fusion gene in non-small cell lung cancerAbstractBackground: Lung cancer continues to be the first incidence and leading cause of cancer death worldwide,and non-small cell lung cancer is the most common type of lung cancer.Currently,molecular targeted therapy is the most promising development part in the treatment of NSCLC.In recent years,more and more attention has been focused on some new molecular targets in NSCLC,such as echinodem microtubule- associated protein-like4/anaplastic lymphoma kinase(EML4-ALK) fusion gene.Objective: The purpose of this study is a review of the basic structure, clinicopathologic features, detection methods,ALK inhibitor and therapeutic meaning of EML4-ALK fusion gene in NSCLC.Methods: An application to retrieve the CNKI Articles Full-text Database,Pubmed, Springerlink database retrieval system,non-small cell lung cancer;EML4-ALK fusion gene;molecular targeted therapy, separately or jointly retrieval 2007-01-2012-12 literature 150.(2)into the pathogenesis of the criteria:1)basic structure;2) clinicopathologic features;3)detection method;4)ALK inhibitor;5)molecular targeted therapy.The inclusion criteria,the selection of 50 documents, and finally included in the analysis of 41 studies.Results: EML4-ALK fusion gene is a new subtype of NSCLC.It is generated as a result of a small inversion within the short arm of human chromosome 2.It has at least 11 subtypes.EML4-ALK fusion gene positive NSCLC is more common in young male patients with mild smoking/no smoking,adenocarcinoma,K-ras、EGFR wild-type. Commonly used method to detect EML4-ALK is IHC,RT-PCR,FISH and RACE-coupled PCR sequencing,each method has its advantages and disadvantages,some complementary.ALK inhibitor PF(Crizotinib) in the treatment of EML4-ALK-positive NSCLC can achieve better efficacy, it is expecte
