明显上升，这些结果都说明吉马酮能够促进HepG2细胞发生凋亡。（5）通过WesternBlotting检测JAKs/STATs信号通路中p-JAK2和p-STAT3的表达情况，结果表明p-JAK2和p-STAT3的表达量随着药物浓度的增加而下降，说明吉马酮能够能够有效抑制JAK2/STAT3信号通路的异常活化，从而达到抑制肿瘤发生的目的。通过以上结果表明，吉马酮具有显著的抑癌效果，并且副作用小，为肝癌的治疗提供了新的思路。关键词：吉马酮肝癌HepG2细胞细胞周期信号通路AbstractRhizomacurcuma,whichisthedryrootorthedrystemoftheCurcumaphaeocaulis,CurcumakwangsiensisandCurcumawenyujinascribedtotheZingiberaceae,hasthefunctionofbloatingthebrokenblood,reducingswellingandrelievingpain.GermacroneisthemainactiveingredientofthevolatileoiloftheR.curcuma.SomeresearchhasreportedthatthevolatileoiloftheR.curcumahasthefunctionofanti-tumor，anti-inflammatoryandanti-virus,andtheresearchdemonstratedthatthegermacronehasanobvioustherapeuticeffectwithlessmechanismreported.TakinghumanHepG2celllineasthetestmaterial,thisthesisexplainstheanti-tumormechanismofgermacrone,byusingthemolecularbiologytoolsWesternblotting,RT-PCRandflowcytometry.Themainresultsinclude：（1）GermacroneinhibitedhumanHepG2cellline’sproliferationinadose-andtime-dependentmanner.MTTassayshowedthatthehigherthedrugconcentration,thestrongertheinhibitiononcellgrowth.Inadditiontherearenosignificantgrowth-inhibitoryeffectdifferencesbetweenincubationwithgermacronefor24hand48h,andnotime-depententinhibitionwasobserved.TheIC50was160μM.Therewasnostatisticaldifferencewhentreatedwithgermacronemorethan160μM.（2）GermacroneinducedG2/Marrestincellcycleprogress.TheflowcytometryassayshowedthatgermacroneinducedanobviousG2/Marrest.WesternblottingandRT-PCRassaysindicatedthatgermacroneinhibitedCyclinB1andCDK1whicharethekeygenesoftheG2/Mphase.ItmeansthatGermacroneinhibitedtheproliferationoftheHepG2cellthroughregulatingtheCyclinB1andCDK1.（3）Expressionlevelsofp53,Bax,Bcl-2weredetectedbywesternblotting.Theresultsshowedthatp53andBaxwereup-regulatedbygermacrone,whileBcl-2wasdown-regulated.（4）CellswerestainedwithHoechst33258andtheresultsshowedthatthebluelightspotweregettingmorewithgermacronetreatment.AlsothecellswerestainedwithPI/AnnexinVwiththeresultsthatthepercentageoflivingcellswasgettin
