ExpressionofIL-6incerebralcortexofmodelratswithfocalcorticaldevelopmentMajor:NeurosurgeryPostgraduate:SUNJieSupervisor:LIUchuangxi,GAOFangyou[Abstract]Objective:ToinvestigateliquidnitrogeninjuryinducesfocalcorticaldisordercerebralcortexinratsexpressionofInflammatorycytokine-6,IL-6.Methods:ItadoptliquidnitrogenfreezedamagedfrontalcortexdamageinnewbornSDratfrontalcortextoestablishfocalcorticaldevelopmentaldisorders(F-MCD)model,usingbehavioralobservationandHEstainingmethodtoevaluatethesuccessofthemodelusedforsubsequentexperiments.3,7,28and60daysafterModeling,itweredrawnmaterialsandthecollectionofanimalspecimens,usingimmunochemicalmethodstoobservethespecificdistributionofIL-6intheratsfrontallobelesionscortexF-MCDmodel.Then,itextractRNAsamplesandprotein,usingRT-PCRandWesternblotdetectionofIL-6intheratsfrontallobelesionscortexF-MCDmodelofchangesingeneexpressionandproteinlevel.Results:LiquidnitrogeninjuredSDratcorticalcaninducetheformationofpathologicalchangesinthemicrobraingyrus,corticaldisorders.3,7,28and60daysafterthepreparationofratsmodelF-MCDcomparedwiththecontrolgroup,IL-6neuronsandastrocytesinthefrontalcortexlesionmodelF-MCDratswereabundantlyexpressed.IL-6expressionatthemRNAlevelwassignificantlyincreased,andtherearesignificantdifferencesbetweenthetwogroups(p0.05).IL6expressionattheproteinlevelwassignificantlyincreased,andtherearesignificantdifferences(p0.05)betweenthetwogroups.Conclusion:1.Withthemethodofusingliquidnitrogeninjuryinducesfocalcorticaldisordercerebralcortex,itsuccessfullyproducethepathologicalandhistologicalanimalmodelsthatsimilartohuman’sFCD.2.Inthedevelopingrats’F-MCDmodel,thelocalfocusareaIL6-expressedonthemRNAandproteinlevelsweresignificantlyup-regulatedandhighlyexpressedinneuronsandastrocytes,suggestingthatIL-6signalparticipationtransductionmaybeanimportantreasonforcorticaldevelopmentaldisordersinducedepilepticmechanism.[KeyWords]inflammatorycytokine-6，liquidnitrogenlesions，FocalcorticaldysplasiaIII目录原创性声明和版权使用授权书??????????????Ⅰ中文摘要???????????????
