OMEGAcompanyofUnitedStates.Primersweredesignedaftersearchingoverthegenebank.ThecodingregionofthegeneofCTLA-4、VDR、MIC-AwasamplifiedbyPCR.AmplifiedPCRproductswereevaluatedby2%agarosegelelectrophoresisandethidiumbromidestaining，wereobservedandtookphotographswithultra-violettransmissionanalysator.TheDNAofCTLA-4、VDRwasidentifiedwithrestrictionenzymedigestion.Digestedproductswereevaluatedby2%agarosegelelectrophoresisandethidiumbromidestaining，wereobservedandtookphotographswithultra-violettransmissionanalysator.AmplifiedPCRproductsofMIC-Agenewereanalyzedbydenaturingpolyacrylamidegelelectrophoresis(PAGE)andsilverstaining，wereobservedandtookphotographswithultra-violettransmissionanalysator.【Results】DNAwassuccessfullyextractedfromblood.ThecodingregionofthegeneofCTLA-4、VDR、MIC-AwasamplifiedbyPCR.TheDNAofCTLA-4、VDRwasidentifiedwithrestrictionenzymedigestion.Digestedproductswereevaluatedby2%agarosegelelectrophoresisandethidiumbromidestaining.AmplifiedPCRproductsofMIC-Agenewereanalyzedbydenaturingpolyacrylamidegelelectrophoresis(PAGE)andsilverstaining.AhighlysignificantincreaseinthefrequencyoftheGallelewasseeninpatientswithLADAcomparedwithcontrols（P＜0.01）.ThisreflectedanincreaseintheGGgenotypeinpatients（P=1.3×10-2)andasignificantdecreaseintheAAgenotype（P＜0.01）.ThefrequencyoftheGallelewassignificantlyhigherinpatientswithLADAthanintype2diabetespatients（P＜0.01）.ThisreflectedanincreaseintheGGgenotypeinpatients（P=3.4×10-2)andasignificantdecreaseintheAAgenotype（P＜0.01).AhighlysignificantincreaseinthefrequencyoftheBallelewasseeninT2DMpatientscomparedwithcontrols（P=2.7×10-2）.ThisreflectedanincreaseintheBbgenotypeinpatients（P=1.6×10-2）andasignificantdecreaseinthebbgenotype（P=1.6×10-2）.TheBborbbfrequencywasnotsignificantlyhigherinpatientswithLADAthaninthecontrolgroup（P＞0.05）,andsowastheBallele.AhighlysignificantincreaseinthefrequencyoftheA5.1allelewasseeninpatientswithLADAcomparedwithcontrols(P＜0.05).TheA5.1allelefrequencywasnotsignificantlyhigherinpatientswithT2DMthaninthecontrolgroup（P＞0
