ABSTRACTIntestineisthemajororganofnutrientsdigestionandabsorptioninpeopleandotheranimals,intestinalmucosahasanimportantbarrierfunction.Mechanicalbarrieristhebasicstructureoftheintestinalbarrierandisconstitutedbyintactintestinalepithelialcellscloselypackedtogetheraswellastightjunctionsbetweentwoadjacentepithelialcells.Tightjunctionsasaselectivepermeabilitybarrier,playanimportantroleinmaintainingbodyhealthespeciallyforthehealthydevelopmentofinfantsafterweaning.Itmayallowsomenutrients,electrolytesandwatermoleculestogothroughtheintestinalmucosa,preventtheintestinepathogenicmicroorganisms,bacteriaandvirusesthroughtheintestinalmucosatoreachotherpartsofthebody.Pigsaregoodmodelforstudyofthemetabolismofvariousnutrients,sincethephysiologicalcharacteristicsofpigsandhumanareverysimilar.Soweselectedpigsasexperimentalanimalmodelsinthisstudytoinvestigatetheeffectofporcineintestinalbarrierfunctionafterdietarytryptophan.Inthefirstexperiment,healthyweaning-piglets(21)wererandomlydividedintothreegroupsaccordingtobodyweight:thecontrolgroup,0.1%Tryptophangroupand0.2%Trpgroup.Thecontrolgroupwasfedthebasaldiet,0.1%Trpgroupwasfedbasaldietplus0.1%Trp,0.2%Trpgroupwasfedthebasaldietplus0.2%Trp,trialperiodis14days.Theresultsshowedthat:(1)Trpincreased(P0.05)thedailyfeedintakeandweightgain,reduced(P0.05)feedconversioninweaning-piglets;(2)Trpelevated(P0.05)diamineoxidaseactivityandendotoxincontentinserum;(3)BasedonileumgobletcellsstaininginjejunumandileumandmorphologyHEstaininginjejunum,wefoundthattrpreduced(P0.05)thenumberofgobletcellsintheintestinalmucosaandcausedpartialdestructionofintestinalvilli;(4)Throughtheimmunohistochemistry,PCRandwesternblottinganalysised,wefoundthattrpreduced(P0.05)theexpressionoftightjunctionproteinZO-1,occludinandclaudin-1.Insummary,tryptophancouldelevatediamineoxidaseactivityandendotoxincontentinserumandreducetheexpressionoftightjunctionproteinsZO-1,occludinandclaudin-1,thusincreasedintestinalpermeabilityinweaning-piglets.Inthesecondexperiment,healthygrowing-pigs(96
