The The Mature Embryo Culture and Rapid Propagation System Estabfishment of ／r／s germanica L． Name：Jia Hongshan Major：Landscape Plant and Ornamental Horticulture Supervisor：Xiao Jianzhong Yin Xinyan Abstract ／r／s germaniea L．，perennial flowers of lfidaceae L．，was the model ofIris．There were dormancy in their seeds which led to low germination rate．The ramet needed a large number of reproductive materials and there were many undesirable phenomena such as characters degeneration after multiples．Stem apex、rhizome and flower organs were mostly used as explants of tissue culture with problems SUch as di伍cult disinfect．10W induction rate and greatly maternal plant damaging．The embryo culture could effectively break the seed dormancy and increase the germination rate and the range of variation of Germplasm． So，The embryo culture was an effective and rapid propagation technology of／r／s germanica． The mature embryos of／r／s germanica No．1(‘Indian chiefX‘Baiyulan’),No．8 (‘Baiyulan’X‘Indian chief)and No．9(‘Indian chiefo)were used as explants to study their vitro rapid propagation technologies．The author explored different disinfection methods and the best disinfection time，screened the optimum germination medium、 subculture medium and rooting culture medium，determined the best hardening-off time and transplanting media of tissue culture seedlings．The aim of this study Was to establish a stable embryo culture rapid propagation system．The main results were as follows： (1)The optimum seed disinfection treatment ofIris germanica No．1，No．8 and No．9 were 75％alcohol sterilizing l Omin and 0．1％HgCl2 sterilizing 8min．The optimum secondary disinfection of No．1 Was 75％alcohol sterilizing 2s，No．8 and No．9 were 75％ alcohol sterilizing 5s．If the disinfection time Was too long，the damage to embryo increased，which was not conducive to the germination of embryos． (2)The optimum germination medium of ／r／s germanica No．1 was MS+6-BA2．0mg／L+IBA0．5mg／L，No．8 and No．9 were MS+6一BAl．0mg／L+IBAl．0mg／L
