结直肠癌RHOA、RHOC基因表达及临床意义 结直肠癌RHOA、RHOC基因表达及临床意义 中文摘要 目的：应用实时荧光定量逆转录聚合酶链反应(real time nuorcSceIlt quaIltitive I盯．PCR)的方法研究结直肠癌RhoA、RhoC表达与临床病理学的相关性，探讨结直 肠癌发生、发展以及转移的分子生物学机制，为结直肠癌的早期发现、早期诊治提 供新的途径。 方法：标本取自青岛大学医学院附属医院普外科手术患者，共分五组，包括： 伴肝转移结直肠癌组、伴淋巴结转移结直肠癌组、不伴转移结直肠癌组、癌旁组织 组、结直肠正常黏膜组；恶性肿瘤标本并按组织学分级和临床病理分期进行分组。 采用实时荧光定量逆转录聚合酶链反应的方法测定标本中RhoA、RhoC mRNA的表 达情况，对实验数据采用t检验和方差分析进行分析比较。 结果：RhoA、RhoC mRNA的表达量在不同临床分期之间(P0．05)，肿瘤 组织学分级之间(PO．05)，淋巴结转移有无转移之间(P0．05)均有显著差异性。 而与年龄、性别、肿瘤部位无关(PO．05)。分期越晚，分化越低，伴淋巴结、肝脏 转移时，RhoA、RhoC mRNA的表达量越高；分期越早，分化越高，无淋巴结及肝 脏转移时，RhoA、RhoC mRNA的表达量越低。 结论：RhoA、RhoCmRNA的定量表达与结直肠癌的临床分期、组织学类型及转 移密切相关；RhoA、RhoCmRNA的定量表达可作为结直肠癌恶性程度的判断指标。 硕士研究生：刘延军(普通外科学) 指导教师：王海波副教授 关键词：结直肠癌； RhoA, RhoC, 荧光定量PCR Expression Expression and clinical significance of RhoA and RhoC in colorectal carcinoma ABSTRACT objective：Using real time fluorescent quantitive RT-PCR to find the correlations of the expression of RhoA and RhoC with clinical pathology of colorectal carcinoma．To explore the molecular biologic mechanism of the occurrence,development and metastasis of colorectal carcinoma．New pathway for earlier detection，diagnosis and treatment of colorectal carcinoma carl be provided． Methods：Samples which were obtained from patients in General Surgery Department of Affiliated Hospital of Medical College Qingdao University were divided into five groups which include the group of coloreetal carcinoma with hepatic metastasis、 the group of colorectal carcinoma with metastasis of lymph node、the group of colorectal carcinoma without metastasis、the group of tissue side carcinoma、the group of normal colorectal mucosa；while the samples of malignant tumor were also divided into several groups through histolo百C classes and clinical pathologic stages．To detect the expression of mRNA of RhoA、RhoC with real time fluorescent quantitive RT-PCR．The experimental data Was analyzed with t test and variance analysis． Results：There are significant deviation of the expressed quantity of mRNA of RhoA、RhoC in d
