夏龙方对人胃癌SGC-7901细胞粘附和侵袭影响 刘威，沈克平［基金项目 ［基金项目］ 国家自然科学基金；上海市浦东新区中医领军型人才项目（PWZ2008-20-104）；上海市级医院临床科研资源共享平台（SHD ［第一作者］ 刘威，在读硕士，E-mail：lw2008126@163.com ［通讯作者］ ?沈克平，学士，主任医师，从事中医药治疗消化道肿瘤临床与基础研究，E-mail: shen0313@163.com 电话：021153 上海中医药大学附属龙华医院肿瘤五科，上海200032 摘要：目的：观察夏龙方对胃癌细胞SGC-7901粘附、侵袭、移动等转移生物学行为的影响。方法：体外培养SGC-7901细胞，分为夏龙方50μg/ml 、100μg/ml 、200μg/ml （低-中-高剂量）组和对照组，采用Cytoselect 48-Well Cell Adhesion Assay检测胃癌细胞与基质粘附能力、利用Transwell侵袭转移模型评价夏龙方对胃癌细胞侵袭能力的影响、划痕实验观察夏龙方对SGC-7901细胞移动能力的影响。结果：终浓度100μg/ml 、200μg/ml夏龙方能显著降低SGC-7901细胞粘附能力，50μg/ml~ 200μg/ml可抑制SGC-7901细胞侵袭，终浓度50μg/ml夏龙方可显著抑制SGC-7901细胞迁移。结论：夏龙方可降低SGC-7901细胞的粘附能力，侵袭能力，抑制细胞迁移。 关键词：夏龙方；中药复方；胃癌；粘附；侵袭；移动 The effects of Shi-teng-fang on adhesion and migration in SGC-7901 gastric carcinoma cells Shen Ke-Ping, Liu Wei, Hu Bing, Pan Chuan-Fang 5th Department of Oncology, Longhua Hospital, Shanghai University of Traditional Chinese Medicine, Shanghai, 20032, China [Abstract] objective：To investigate the influence of Xia-long-fang on adhesion, invasion and migration in SGC-7901 gastric carcinoma cells. Methods: Human gastric carcinoma SGC-7901 cells were cultured in vitro, and treated with different concentration Xia-long-fang. (0μg/ml~200μg/ml ). Cell adhesion was detected with cytoselect 48-well cell adhesion assay. Cell invasion was analyzed with Transwell invasion assay. Cell migration was detected with scratch wound healing assay. Results：50μg/ml~200μg/ml Xia-long-fang significantly inhibited the invasive capacities of SGC-7901 cells and inhibited cell adhesion at 100μg/ml ~ 200μg/ml. Wound healing assays showed a significant reduction in the migrated cells after treated for 24h or 48h at 50μg/ml Xia-long-fang. Conclusions: Xia-long-fang significantly decreased the adhesion and invasive capacities of SGC-7901 cell lines and inhibited cell migration.
