Correspondent: XU Shu-kun, Professor, E-mail: xushukun46 @ 126. com) Abstract: Gold nanoparticles w ith 14 nm , 19 nm and 40 nm in diameter were synthesized through citrate sodium reduction method, and all of them w ere labeled w ith BSA. The influencing factors on the conjugation of colloidal gold to BSA were discussed. M igrations of those gold nanoparticles and their Au-BSA conjugates in agarose-gel electrophoresis w ere studied. Experimental results indicated that pH alue, ion concentration and protein addition all ha e influences on the conjugation of colloidal gold to BSA. The optimal conditions for BSA labeling are salt-free or ery low salt concentration and pH alue= 5. 8 and especially BSA addition isn t less than the minimal stabilizing protein concentration. Migratory beha ior of gold nanoparticles and Au-BSA conjugates in the applied electric field is similar to that of rocket immuno-electrophoresis, w here the migration speed decreases w ith increasing gold nanoparticles size.
