香菇多醣及其水解產物生理活性之探討 Studies of Physiological Activities of Lentinan and its Hydrolysates 研究生:蔡侑蓁 指導教授:廖遠東Ean-Tun Liaw 【摘要】 香茄多醣為β-(1→3)(1→6)-葡聚醣。本研究包含體外試驗香菇多醣水解產物之益生物質特性、以動物模式探討香茄多醣及其水解產物之降血脂功效,以及評估兩者對體外癌細胞生長之影響三大部分。實驗結果歸納如下: 香菇多醣水解產物於體外促進益生菌生長試驗,3.5小時水解產物具最佳益生物質特性,促進L. acidophilus生長的能力為空白對照組之800至2000倍,市售寡醣之30至1000倍;而促進B. bifidum生長之能力分別的空白組及市售寡醣產品之700至900倍和35至900倍。香菇多醣及香菇寡醣抑制癌細胞生長試驗,發現香菇多醣(0~1,000μg/mL)或香菇寡醣(0~100%)無法誘發體外培養細胞死亡,而MTT assay發現可降低Hep G2、Siha、MCF-7及HBL-l00存活率,且具劑量依存性,正常細胞株HBL-100之耐受性最高。香菇多醣對細胞株存活率之影響大於香姑寡醣。再者,短時間共同培養(24或48小時)之抑制效果大於長時間(72小時)培養。 香菇多醣及香菇寡醣於體內之降血脂功效,實驗飲食8過後攝食低量香姑多醣之倉鼠三酸甘油酯顯著(p0.05)高於控制組;攝食香菇多醣可明顯(p0.05)降低膽固醇;攝食香菇寡醣則顯著(p0.05)提昇高密度脂蛋白膽固醇。而攝取香菇多醣及香菇寡醣可顯著增加(p0.05)倉鼠肝臟重量;香菇多醣對盲腸有顯著增重(p0.05)效果,而香菇寡醣無。外觀及病理解剖結果顯示,攝食香菇多醣較香菇寡醣更能防止脂肪肝發生,此效果具劑量依存性。反之,攝食香菇寡醣之肝腫脹比例較低。足量香菇寡醣有助於清除血管中脂質蓄積,低童則無法發揮效用;肝臟脂肪蓄積改善情形具香菇多醣劑量依存性,香菇寡醣則無法清除肝臟脂肪。 關鍵字:香菇多醣、香菇寡醣、生理活性 【Abstract】 Lentinan, a β-(1→3)((1→6)-glucan, was isolated from an edible mushroom, Lenrinus edodes. The aims of the present study were to extract lentinan from local species of Lentinus edodes, prepare the partial hydrolysates of lentinan, and examined the potential physiological properties of both of them. The results were summarized as follows: In vitro, the results of prebiotic test demonstrated that the effect of various hydrolysates on the growth of L. acidophilus was 880 to 2,000 folds faster than that of blank where without any supplement under defined conditions. The results also indicated that the hydrolysates showed 30 to 1,000 times higher rate in proliferation on L. acidophilus than those of commercial oligosaccharides. However, similar tendency with lower stimulation ability was observed in B. bifidum compared to L. acidophilus. Lentinan and lentioligosaccharide show stronger growth inhibition on Siha, Hep G2, and MCF-7 cancer cell lines than HBL-100 cell, which was normal cell lines, and the inhibitory effects were dosage-dependent.
