PAGE PAGE II 常州工程职业技术学院 毕业设计报告（论文） 系 别： 制药与生物工程技术系 课题名称： 蔬菜中的亚硝酸盐含量测定 摘 要 近几年来我国人口中患消化系统癌症的人数明显增多，根据学者的研究发现，造成这种现象的原因与我国人口摄入过多的硝酸盐和亚硝酸盐是分不开的。工业、农业的发展和生活燃烧导致了地区和全球氮循环的混乱,亚硝酸盐的渗透还引起地下水污染,威胁着人类的健康。尤其是化学氮肥的广泛使用， 蔬菜中的硝酸盐残留量过高。 人体持续摄入少量的硝酸盐会引起消化不良、精神抑郁和头痛,当摄入0.3-0.5g亚硝酸盐即可引起中毒, 3克可致死。另外亚硝酸盐能够透过胎盘进入胎儿体内,人体血液中亚硝酸钠浓度为 0.4mg/kg～200mg/kg时会产生毒性,孕妇食用过量的亚硝酸钠可导致胎儿畸形,特别是六个月以内的婴儿。联合国粮农组织和世界卫生组织规定硝酸盐和亚硝酸盐的每日允许摄入量： KNO3 或 NaNO3 0.5mg/kg，KNO2或 NaNO2 0.2mg/kg。 为了更加健康的饮食习惯，本文开始研究蔬菜中的亚硝酸盐含量。针对亚硝酸盐的测定，本文借鉴前人的经验设计出一套合理可行的分光光度法测定蔬菜中亚硝酸盐的方案，该方案的线性范围为0 mg/L～0.3 mg/L，线性相关系数r=0.999 x，检出限为0.14 mg/kg，回收率为94.61%～104%的亚硝酸钠含量。分别测出芹菜、西红柿、大蒜在常规条件下的亚硝酸盐含量，分别对这三种蔬菜在不同浸泡时间、不同煮沸时间及不同放置时间下的亚硝酸盐含量进行检测。不难发现减少亚硝酸盐的摄入，科学的蔬菜处理法是要浸泡至少20min，煮蔬菜时尽量减少煮沸的时间，煮熟的蔬菜隔夜的不宜食用。另外还可以从该实验中看出叶菜类蔬菜中含有的亚硝酸盐含量要高于茄果类蔬菜和鳞茎类蔬菜。 关键词：分光光度法 蔬菜 亚硝酸盐 探究 Abstract In recent years Chinas population with the digestive system in the number of cancer significantly increased, according to the research of the find, cause the reasons of this phenomenon and our country population excessive intake of nitrate and nitrite is not divided.
