Highlevel expression and purification of porcine pepsinogen A in pichia pastoris

Abstract: A system ofp ich iap astor is for high- levelexpression ofporcinepepsin, one kind of mi por
tant protease both in industrial application and academ ic research, was reported in this paper. The
DNA encoding pepsinogen A was inserted intoXhol andXba sitesof the vector pGAPZ. Afterp.
p astoris transformation, clonieswere screened for expression ofpepsinogen based on enzym e activity of
its active form, pepsin. The recombinant enzym ewas purified by DEAE ion- exchange and Sephacryl
S- 200 gel filtration chromatography. Homogeneitywas confirmed through SDS- PAGE, the specific
activity ofpepsinwas about 52 800U /g; the expression levelwas 5 250U /L. The results indicated
that the porcine pepsinogen was high- level expressed, and it could be efficiently purified.
