During formation, autophagosomes sequester cytoplasmic contents and often include mitochondria. The presence of mitochondrial material inside a vacuole is indicative of a compartment representing a bona fide autophagosome. Irgm1- induced vacuoles were positive for Mitotracker Red (MT) , with a sevenfold increase in MT+ cells among GFP-Irgm1–transfected cells (65 ± 5%) compared to controls (9.4 ±3.5%; P ＜0.01) .
