Summary

Anin-situhybridization(ISH)procedurewasdevelopedforthedetectionofCryptosporidiumsp.inparafﬁn

wax-embeddedtissueswithadigoxigenin-labelledprobetargetingthe18SrRNA.Thistechniquewasused

inadditiontotraditionalmethods,suchashaematoxylinandeosinstaining,periodicacid-Schiffreaction,

transmissionelectronmicroscopyandthepolymerasechainreaction,toexaminethebursaofFabricius

(BF),conjunctivaandothertissuesfrom20domesticgeeseaged16–36daysforthepresenceof

cryptosporidia.Positivesignalswerefoundtoamoderateormarkedextentinbothconjunctivalsamples

(89%)andBFsamples(88%)butnotinothertissues.SequencingofthePCRampliﬁcationproduct

revealedidentitywithCryptosporidiumbaileyi.Theinfectedgeeseshowednoclinicalsignsandonlyscanty

histologicallesions.Theseresultsconﬁrmreportsshowingthatyoungwaterfowlareespeciallyvulnerable

tocryptosporidiuminfectionandindicatethattheBFandconjunctivaarethepref
