甘露糖结合凝集素论文：MBL基因多态性与新疆汉族结核病易感性的相关性研究 【中文摘要】探讨甘露糖结合凝集素(mannose-binding protein,MBL)基因多态性与新疆汉族人群结核病(TB)发生发展的关联。方法：采用病例对照研究方法和序列特异性引物-聚合酶链反应(polymerase chain reaction-specific sequence primers PCR-SSP)技术对231例新疆塔城地区和石河子地区的汉族肺结核患者及226例健康志愿者的MBL基因H/L、P/Q和A/B三个多态性位点进行基因及单倍体分型,比较各等位基因及单倍体型的频率(f),并计算其优势比(OR)。统计学分析：应用SPSS13.0统计软件采用χ2检验或Fisher’s精确检验,研究不同等位基因及单倍体型与新疆汉族人群结核病易感性的关联。结果：对MBL-AB位点等位基因进行检测,发现新疆汉族人群结核病例组的AA基因型频率为74.03%,明显低于健康对照组中的AA基因型频率82.30%,两者经检验,有统计学意义(χ2=4.576,P=0.0320.05)。结论：1. MBL-AA基因型与新疆汉族结核病显著负相关,其可能是新疆汉族结核病的抵抗基因。2. MBL-AB基因型与新疆汉族结核病显著相关,其可能是新疆汉族结核病的易感基因。3. MBL-H/L、P/Q位点的基因型及单倍体型与新疆汉族结核病无明显关联。 【英文摘要】:To explore the association between the genetic polymorphism of MBL and susceptibility to pulmonary tuberculosis in Xinjiang Han population.Methods:A case-control study was conducted,chosing 231 XinJiang Han patients with tuberculosis from Ta and Shihezi cities to be genotyped for H/L、P/Q and A/B SNPs in MBL gene by using the sequence-sp-ecific polymerase chain reaction (PCR-SSP).Gene frequency (f) and odds ratio (OR) were calculated and c-ompared with those of 226 healthy individuals from the same areas.Using the SPSS 13.0 to analyze the difference of the frequency of MBL three alleles and haplotype between the two groups.Chosing Chi-squar-e test or Fisher抯 exact test to analysis the association between alleles and tuberculosis.Result:The frequency of MBL-AA homozygote genotype in the group with tuberculosis disease is 74.03%,and it is significantly lower than 82.30% in the group of control in XinJiang hans.The significant difference is remained after the two group being tested (x2=4.576, P=0.0320.05).Conclusions:1.The results showed that MBL-AA homozygote genotype is probably a protective gene in tuberculosis for Xinjiang Han population;2. The results showed that MBL-AB homozygote genotype may be a susceptible gene in tuberculosis for Xinjiang Han population;3.
