 The Effect of Atorvastatin on HDL Subfraction Phenotype in Patients with Atherosclerosis# XU Ruixia, GUO Yuanlin, LI Xiaolin, LI Sha, LI Jianjun** 5 10 15 20 25 30 35 40 (State Key Laboratory of Cardiovascular Disease, Fu Wai Hospital, National Center for Cardiovascular Disease, Chinese Academy of Medical Sciences and Peking Union Medical College) Abstract: Background and Objective Although statins (HMG-CoA reductase inhibitors ) have been shown to modestly raise or not alter HDL-cholesterol and significantly reduce LDL-cholesterol in patients with coronary artery diseases, their effects on HDL subfractions and LDL subfractions has not yet been fully explored. The objective of this study was to investigate the short-term impact of low-dose atorvastatin on HDL subfractions and LDL subfractions in humans. Methods In this randomized study, data from 52 subjects were analyzed. 37 patients with atherosclerosis were randomized to atorvastatin 10mg/day or 20 mg/dayand 15 healthy subjects without therapy were controls for 8 weeks. At baseline and after 8 weeks of treatment, the lipid profile and the lipoprotein subfractions were determined by Lipoprint system. Results Atorvastatin 20 mg/day for 8 weeks treatment increased the cholesterol concentration of large HDL particles and decreased the cholesterol concentration of small HDL particles without altering serum HDL-C level in patients with atherosclerosis. Atorvastatin treatment (10 and 20 mg/day) decreased LDL-C by reducing the cholesterol concentration of all LDL subfractions. Conclusion The low-dose atorvastatin changed HDL subfractions phenotype and reduced the cholesterol concentration of all LDL subfractions. The measurement of HDL subfractions can be used to assess the clinical benefits of HDL-targeted therapies, including statin treatment.
