The effect of diazepam on seisure induced by metrazol Purpose To observe the antiepileptic effect of diazepam against convulsions induced by metrazol Principle Metrazol is a central stimulant drug, which can stimulate brain and spinal cord, resulting in clonic convulsion and tetanic convulsion successively. The action site of metrazol may be located in brain stem and cerebrum. The action mechanism of metrazol concerns the increase of brain neuron cell’s permeability to K+ and enhancing the concentration of extracellular K +, resulting in cell membrane depolarization and increasing neuron excitability and facilitation of stimulant synapse. At the threshold dose ,metrazol result in forelimb and head convulsion, but it doesn’t produce any influence on the righting reflex, so this action is called the metrazol minor seizure model(MMS). Large dose of metrazol lead to systemic clonic convulsion , tonic convulsion and even death Successively.This action is called Metrazol maximal seizure test(MMS, major epilepsy). Materials Animals: mouse. Drugs:0.02% diazepam, Metrazol, normal saline. Equipments: syringes Methods Six mice are divided into 2 groups randomly(each group has3 mice), NS group and Diazepam group. Diazepam group is administered with diazepam 0.1ml/10g BW intraperitoneally, NS group is injected normal saline 0.1ml/10g BW intraperitoneally. 30 minutes later,all animals are treated with metrazol (0.2ml/10 g) intraperitoneally. Observe the numbers of convulsant animal and ratio of convulsion within 30 minutes. The characteristics of mice’s maximal seizure convulsion: clonus convulsion 5-15 minutes after metrazol administration or excitability jump, then forelimb flection, hindlimb stiffness, Hindlimb stiffness as convulsive indicator.
