National Medical Frontiers of China, Sep 2010, Vol.5 No.18 中国医疗前沿 2010年9月 第5卷 第18期 基础研究 19 丙氨瑞林对子宫内膜癌裸鼠移植瘤的作用及机制 董美娥 张三元 【摘要】 目的 探讨促性腺激素释放激素 (GnRH) 类似物丙氨瑞林对人子宫内膜癌裸鼠皮下移植瘤的作用机制及对P16基因 表达的影响。方法 体外培养人子宫内膜癌 Hec-1B细胞，建立子宫内膜癌动物模型。造模成功后分别用不同剂量的丙氨瑞林 进行干预，干预过程中严密观察肿瘤生长及裸鼠情况，测量肿瘤体积,绘制肿瘤生长曲线。6周后处死裸鼠，完整取出移植瘤 组织并称取瘤重，计算抑瘤率，免疫组化法(PV)检测瘤组织中P16基因的表达。结果 当丙氨瑞林浓度为15μg/kg时，癌细胞即 受到抑制，抑瘤率为 20.79% ；当其浓度为30μg/kg时，抑瘤率为 34.34% ；浓度为60μg/kg时，抑瘤率为 39.23%。与对照组比较差 异有统计学意义(P ＜0.05)。不同浓度的丙氨瑞林干预6周后，瘤组织中P16的表达有不同程度的上升，与对照组相比，差异有 统计学意义(P ＜0.05)。结论 丙氨瑞林可抑制裸鼠移植瘤生长，且呈剂量依赖关系，其抑制作用的分子机制可能与P16基因的 表达有关。 【关键词】 子宫内膜癌 ；促性腺激素类似物 ；丙氨瑞林 ；裸鼠 ；P16 doi:10.3969/j.issn.1673-5552.2010.18.0009 【中图分类号】R737.33 ；R364.3 【文献标识码】A 【文章编号】1673-5552(2010)18-0019-02 【Abstract】Objective To evaluate the effects of gonadotropin releasing hormone analogue (GnRH-a) Alarelin on endometrial cancer xenograft in nude mice and the expression of p16. Methods Human endometrial carcinoma hec-1-b cells were cultured in vitra. After establishing the model of human endometrial cancer xenograft in nude mice, the model mice were injected subcutaneously with diffrent concentrations of Alarelin. During the period of injection with Alarelin，we measured the tumor size, observed the condition of the mice tumors and drawed the growth curve of the tumors. After 6 weeks we executed the nude mice, dislodged the tumors completely, measured the weight of tumors, calculated the inhibition rate of the tumor and detected the expression of P16 gene by immunohistochemistry (PV). Results The growth of tumor was inhibited when the 1owest Alarelin concentration 15μg/kg was given, the inhibitive rate of the tumor was 20.79%. When Alarelin concentration was 30μg/kg, the inhibitive rate increased to 34.34%.
