胚胎心肌细胞移植到急性梗死心肌区的实验研究_医学论文 胚胎心肌细胞移植到急性梗死心肌区的实验研究_医学论文 作者：郑奇军　蔡振杰　俞世强　杨光　胡军　胡巧侠 【关键词】 急性梗死 关键词: 心肌/细胞学急性梗死新生血管化，病理性移植 摘 要:目的 观察移植的胚胎心肌细胞在成年兔急性梗死心肌组织内的生长发育过程和其与宿主心肌组织的相互作用，以及对周围心肌血运的影响. 方法 将培养的兔胚胎心肌细胞，经DAPI标记后，直接注入成年兔的急性梗死心肌组织内（通过结扎左冠状动脉前降支建立急性心肌梗死模型），同时设立对照组.4wk后处死动物，进行组织学和电镜观察. 结果 4wk后的心肌组织切片中，移植心肌细胞与宿主心肌细胞之间形成闰盘，其排列方向与宿主心肌肌纤维方向平行，同时，与对照组相比，心肌梗死区面积减少及移植区血管密度增高（0.05）. 结论 移植的胚胎心肌细胞在受体急性梗死心肌内可以继续生长发育，逐渐分化成熟，从而稳定病损的心肌结构，改善心脏的收缩功能，同时，对改善周围血供起到积极作用. 　　Keywords:myocardium/cytologymyocardial infarctionneovascularization，pathologictransplantation 　　Abstract:AIM To investigate the interaction between im-planted fetal cardiomyocytes and host cardiomyocytes and the angiogenesis effects of implanted fetal cardiomyocytes on the host cardiomyocytes and the development of fetal cardiomy-ocytes implanted into adult acute infarction myocardium.METHODS After labelled with DAPI，cultured rabbit fetal cardiomyocytes were implanted into the actue myocardial in-farction site of adult rabbits via direct injection.The myocar-dial infarction model was established by ligating the left ante-rior decending artery.Specimens were harvested4weeks af-ter the cell implantation.The results were observed by histo-logic methods and electronic microscopy examination.RE┐SULTS Intercalated disks were observed between the graft-ed and the host cardiomyocytes，with the implanted car-diomyocytes parallel to the host myocardium in an aligning di-rection.The area of myocardial infaction decreased and the vascular density of implantationsites increased compared with those of the control groups（0.05）.CONCLUSION The fetal cardiomyocytes，after being implanted into the acute myocardial infarction，can differentiate into mature car-diomyocytes in the host heart，which not only repair the structure of the damaged cardiomyocytes and improve the systolic function of the host heart，but also induce angiogene-sis in the core of the infarct. 　　0 引言 　　将体外培养的兔胚胎心肌细胞直接移植到成年兔急性梗死区的心肌内，分别观察研究其在受体心肌组织内的生长发育和与宿主心肌细胞的
