Institute of Forestry Science ofOrdos Inner Mongolia,Ordos 017000,China) Abstract :The paper asbasedon traditional soilmicroorganismisolated culturemethod, it made gropingresearch to affect bacteria, fungiand actinomycetes isolated effects several important factorsselected several important influencing fac2 tors respectively though the direct cross test and single factor test.Thegoal is to discuss and solve present situation that isolation and culture of microorganism in soil is difficult through the practical and feasible means.The experiment has conducted theseparation characteristicsresearch ofbacteria,funguses and actinomyces through culturemediumscreening, the different diameter ofglass beadhitting,as ell as chemistry dispersing agent and theduration ofoscillation and so on four factorsorthogonal designs.At the sametime,there is t ofactors singlefactor experiments includingthe heat treatment temperature of soil sample and the time of heat treatment on isolation ofactinomyces.The result ofResearching indicated that bacteriacs separating optimal combination is SABA PYJ+ Bigbeading ( 5= 5- 7mm,55 gra
