Not Intended for Diagnostic or Clinical Procedures. INTRODUCTION Beta-glucosidase is an enzyme that acts on the beta-1,4 linkage in two glucose molecules. It is an exocellulase that specifically acts upon a variety of beta-D-glycoside substrates. It also catalyzes the hydrolysis of non-reducing terminal residues in beta-D-glucosides to produce glucose. One beta-glucosidase includes lysozymes, which is found in tear secretion to prevent bacterial infection of the eye. It is discovered to cleave the beta-1,4 linkage between N-acetylglucosamine and N-acetylmuramic acid sugars in peptidoglycan in gram-negative bacteria. This assay uses fluorescein digalactoside (FDG), a non-fluorescent galactosidase substrate, which is cleaved by galactosidase to form a highly fluorescent fluorescein that can be read with a fluorescence instrument with FITC filter set. The fluorescein product can be read with fluorescence microplate reader, fluorescence microscope, or flow cytometer with excitation at 490 nm and emission at 525 nm. Higher signals correspond to higher levels of galactosidase, allowing us to distinguish between LacZ+ from LacZ- cells and detect galactosidase conjugates for ELISA assay systems with high sensitivity.
