INTRODUCTION Beta-Hydroxybutyric acid (also known as beta-hydroxybutyrate or 3-hydroxybutyric acid), along with acetoacetate and acetone, are ketone bodies that are produced during ketosis. In humans, beta-hydroxybutyrate is synthesized in the liver from acetyl-CoA and can be used as an energy source by the brain when blood glucose is low during starvation. In clinical settings, patients can have their ketone levels tested via urine or blood samples to indicate ketoacidosis, which can result in metabolic problems. In this assay, beta-hydroxybutyrate undergoes a series of reactions that produces NADH, which can be measured at 340 nm.
