人参单体皂甙Rh2对人喉癌细胞株Hep-2 抗增殖作用的研究 于广久1,康　健1,郭　凤2 (1·锦州医学院附属第一医院耳鼻咽喉-头颈外科,辽宁　锦州　121000; 2·齐齐哈尔市第一医院耳鼻咽喉科,黑龙江　齐齐哈尔　161005) 锦州医学院学报 〔作者简介〕于广久(1977-),男,辽宁省丹东市人,在读硕士研究生,主要研究方向是喉癌的肿瘤学治疗。 【摘要】目的　探讨人参单体皂甙Rh2对体外培养的人喉癌细胞株Hep-2的抗增殖作用及其作用机制。 方法　采用四甲基偶氮唑蓝(MTT)实验、流式细胞技术及免疫组织化学S-ABC法分别检测人参单体皂甙Rh2对喉癌细胞株Hep-2的增殖、细胞周期的影响。结果　(1)人参单体皂甙Rh2可明显抑制Hep-2细胞的生长,并呈剂量、时间依赖性作用。(2)人参单体皂甙Rh2 (30μg/ml)处理细胞24h: G1期细胞由60·09%增加至68·86% (P0·01), S期细胞由18·89%减少至12·30% (P0·01), G2/M期细胞及凋亡细胞无明显变化(P0·05),细胞阻滞于G1期。结论　人参单体皂甙Rh2对人喉癌细胞株Hep-2具有明显的生长抑制作用,并可导致其G1期细胞周期阻滞。G1期细胞周期阻滞可能是人参单体皂甙Rh2抗肿瘤作用的机制之一。 【关键词】人参单体皂甙Rh2 (G-Rh2);喉癌;细胞周期 【中图分类号】　R739·65　　　【文献标识码】　A　　　【文章编号】　1000-5161 (2003) 06-0034-04 Anti-proliferative Effect and the Mechanism of Ginsoniside Rh2on Human Laryngeal Carcinoma Cell Line Hep-2YU Guang-jiu1, KANG Jian1, GUO Feng2 (1·Department of ORL-HNS, the First Affiliated Hospital of inzhou Medical College, Jinzhou 121000 China; 2·Department of ORL,the First Hospital of Qiqiha’er City, Qiqiha’er 161005 China) 【Abstract】　Objective　To study the anti-proliferative effect and the mechanism of Ginsoniside Rh2 (G-Rh2) on human laryngeal carcinoma cell line Hep-2·Methods　MTT assay was used to evaluate the growth inhibition effect of G-Rh2 on human laryngeal carcinoma cell line Hep-2 and the flow cytometry for cell cycle analisis·Results　G-Rh2 significantly inhibited the growth of Hep -2 cells with a dose-time-independent manner·After 24 hours treatment with 30μg/ml G-Rh2, the proportion of cells in G1phase increased from 60·09% to 68·86% (P0·01), while the cells in S phase decreased from 18·89% to 12·30% (P0·01) and there is no significantly change about cells in G2/M phase and apoptotic cells·Conclusions　G-Rh2 inhibited the growth of human laryn- geal carcinoma cell line Hep-2 significantly, arrested cell cycle in G1phase·It is the anti-tumor mechanism for G-Rh2 possibly by arresting cell cycle· 【Key words】　ginsoniside Rh2 (G-Rh2); cell cycle; laryngeal carcinoma 　　人参单体皂甙Rh2 (G-Rh2),为近年来从人参中提取的一种新的有效活性成分。现代医学研究发
