Colchicine induced tetraploids in yellow-flowered cyclamens and their characteristics

ELSEVIER Scientia Horticulturae. 65 ( 1996) 305-3 12 SCIENTIA HOmlCULnJRR Colchicine induced tetraploids in yellow-flowered cyclamens and their characteristics Takejiro Takamura * , Ikuo Miyajima Laboratory of Horticultural Science, Faculty of Agriculture, Kyushu University, Fukuoka 812-81, Japan Accepted 9 February 19% Abstract The possibility of obtaining tetraploids from diploid yellow-flowered cyclamen ‘Kage Yellow’ through colchicine treatment and the effects of polyploidization on the characteristics of yellow- flowered cyclamen were investigated. A Cday treatment of tuber segments with 100 mg 1-l colchicine in vitro yielded two tetraploid plants. The petals of the tetraploids were larger and had a greater ability to accumulate chalcone than those of their diploid relatives. Polyploidization may therefore represent a useful method for the commercial breeding of deeper yellow-flowered cyclamen. Keywords: Cyclamen; Colchicine; Polyploidization; Yellow-flowered, Chalcone 1. Introduction Cyclamen perurn Mill. is the only commercially-important species of Cyclamen. A large number of cultivars with flowers ranging between red, pink, white, reddish-purple and purple have been recorded in this species. Ever since Miyajima et al. (1991) observed an individual specimen with yellow flowers in a population of white-flowered cyclamen ‘Pure White Kage’ (2x), efforts have been directed towards obtaining cultivars with deep-yellow flowers. C. persicum cultivars variously exist at the diploid (2n = 481, tetraploid (2n = 96) and aneu-tetraploid (2n = 90, 92, 94 or 95) levels (Legro, 1959). The tetraploids are autoploids that spontaneously arise from the diploid, and include cultivars with red, pink and purple flowers; tetraploids with yellow flowers were not known prior to this study. * Corresponding author at: Department of Horticulture, Faculty of Agriculture, Kagawa University, Miki-cho, Kagawa 761-07, Japan.
