QUALITY CONTROL PROCEDURES
I INTRODUCTION
Sodium Hippurate Broth is a culture medium for the presumptive identification of group B streptococci on the basis of hippurate
hydrolysis.
II PERFORMANCE TEST PROCEDURE
1. Inoculate representative samples with the cultures listed below.
a. Using sterile pipettes, inoculate tubes with 0.1 mL of 10-1 dilutions of 18- to 24-h Trypticase? Soy Broth cultures.
b. Incubate tubes with loosened caps at 35 ± 2°C in an aerobic atmosphere.
2. Examine tubes after 48 and 72 h for growth.
3. Centrifuge the broth cultures to sediment the growth. Transfer 0.8 mL of the clear supernatant fluid to a clear tube. Add
0.2 mL of ferric chloride reagent prepared as described below. A heavy, cloudy precipitate that persists after 10 min with
shaking is considered a positive reaction. A clearing of the initial precipitate is considered a negative reaction.
4. Expected Results
Organisms ATCC? Recovery Reaction
*Streptococcus agalactiae 4768 Moderate to heavy growth Hippurate hydrolyzed
*Streptococcus pyogenes 19615 Moderate to heavy growth Hippurate not hydrolyzed
*Recommended organism strain for User Quality Control.
III ADDITIONAL QUALITY CONTROL
1. Examine tubes as described under “Product Deterioration.”
2. Visually examine representative tubes to assure that any existing physical defects will not interfere with use.
3. Incubate uninoculated representative tubes aerobically at 20–25°C and 30–35°C and examine after 7 days for microbial
contamination.
