China J Orthop T rauma, August 2000, Vol. 13, No. 8 ? 459 ? 增骨 号影响 、 型胶原基因表达的实验 研究 魏玉玲1 ! 王炳南1 ! 梁克玉1 ! 李文鑫2 ( 1 湖北中医学院附属医院, 湖北 ! 武汉 430061; ! 2 武汉大学生命科学院, 湖北 ! 武汉) ! ! ? 摘要# ! 目 的 ! 从 分 子 水 平 探讨 增 骨 号的 作 用 机 理。方 法 ! 取 120 只 昆 明 小 鼠 造 成 桡 骨 15mm 缺损骨折, 随机分为 2 组: 服 药组 及未 服 药组, 各 60 只, 在 术 后 2 周 和 4 周, 分 别进 行骨 痂 总 R NA 的分离提取, 通过与 、 型胶原质粒探针( pM Col1a1 及 pM Col2a1) 斑点 杂交, 比较 、 型胶 原 mR NA 表达的 变化。结 果 ! 型胶原 mR NA 表达在 术后 2 周 明显小于 4 周, 型胶原 mRNA 表达 在 术后 2 周明显大于 4 周, 提示 型胶原 mRN A 表达代 表骨形成及塑 形的特征标 志, 型胶原 mRN A 表 达代表软骨修复的特征标志。 型胶 原 mRN A 表达 量, 服药 组 大于 未服 药组; 型胶 原 mR NA 表 达 量, 服药组小于未服药组。提示服药组骨形成迅速, 由软骨修复期提早进入骨修复及塑形期。结论 ! 增 骨 号能促进及加快骨的修复。 ? 关键词# ! 骨质疏松症 ! ! 中草药 ! ! 基因表达 Experimental study of the effect of Zenggu No. on expression of type and type collagen gene ! CH( Chinese Herbal) Zenggu No . is one of a serial medicines for t he treat ment of osteo porosis; gene technology is applied in this ex periment for the investigation of its molecular mecha nism. Methods ! M odels of radial fracture with bone defects of 15 mm were pro duced in 120 mice. T he ani mals w ere divided into 2 groups randomly of the treatment and control groups. A t the 2nd w eek after opera tion, total R NA in callus of fracture site of the tw o groups were extracted and were hy bridized w ith probes of pM Col1a1 and pM Col2a1 in dot blot.
