Abstract In order to explore the FCM in the minimal residual disease (MRD) detection of the significance of using the 4-6 group of four-color antibody combinations in order to CD45/SSC gating method, 273 cases of adults and 142 children with B-lineage acute lymphoblastic leukemia ( B-ALL) patients had multi-parameter flow cytometry (FCM) immunophenotyping detection and leukemia-associated immune phenotype (leukemia associated immunophenotyping, LAIP) analysis. The results showed that: According to the expression of CD34 and CD10 characteristics of B-ALL patients can be divided into I-type (CD34 CD10-); Ⅱ-type (CD34 CD10); Ⅲ type (CD34-CD10); Ⅵ-type (CD34-CD10-) . In patients with type Ⅰ and type Ⅱ LAIP positive rates were 100% and 92%, but the type Ⅲ patients LAIP positive rate was only 79.2%, significantly lower than that in patients with type Ⅰ and type Ⅱ. B-ALL patients LAIP overall detection rate of 90%, adults and children was no significant difference. The use of CD34/CD10/CD19/CD45 a group of antibodies, LAIP detection rate of 77.6%. Conclusion: 90% of adults and children with B-ALL leukemia patients LAIP, and therefore suitable for these patients with multi-parameter flow cytometry for MRD monitoring.
