Pump inhibitor on Acinetobacter baumannii-induced mutant fluoroquinolone sensitivity

PAGE \* MERGEFORMAT 15 Pump inhibitor on Acinetobacter baumannii-induced mutant fluoroquinolone sensitivity [Abstract] Objective To study the active efflux pump in Acinetobacter baumannii-induced mutant strains of fluoroquinolone sensitivity. Methods Ciprofloxacin-induced Acinetobacter baumannii mutant screening. Agar double dilution pump inhibitor carbonyl cyanide m-chlorophenylhydrazone (carbonylcyanide  m  chlorophenyl  hydrazone, CCCP) or the presence of reserpine-induced mutation in Acinetobacter baumannii strains to ciprofloxacin, levofloxacin and processing for the Sand Star-sensitive changes. Results CCCP and reserpine can affect the part of the mutant strain of Acinetobacter baumannii on three kinds of fluoroquinolone minimum inhibitory concentration (minimum inhibition concentration, MIC), to enable MIC down to the original value of 1 / 4 of the following pump-positive strains in on the three kinds of fluoroquinolones are sensitive to the presence of the mutant strains. Conclusion increased expression of active efflux pumps may be Acinetobacter baumannii mutant elevated MIC values of one of the reasons [Keywords:] Acinetobacter baumannii; fluoroquinolone; active efflux pump; pump inhibitors ABSTRACT Objective To investigate the role of efflux pump of Acinetobacter baumannii mutant strains against fluoroquinolones. Methods The mutant strains of Acinetobacter baumannii separated from clinical sample were selected by ciprofloxacin. The antibiotic activities of ciprofloxacin, levofloxacin and gatifloxacin were detected by 2  fold agar dilution method with or without efflux pump inhibitors, carbonylcyanide  m  chlorophenyl  hydrazone (CCCP) or reserpine. Results CCCP and reserpine could reduce the minimum inhibition concentration (MIC) to below 1 / 4 of former in some of the mutant strains against fluoroquinolones. Positive efflux pump strains existed in mutant strains which were susceptible to them. Conclusion The excessive expression of
