Using MTT method and the sandwich enzyme-linked immune complexes (ELISA) to detect G-PB and G-BM T-lymphocyte proliferation and IL-4, IFN-γ secretion, and detected by flow cytometry using two kinds of graft T-lymphocyte subsets, dendritic cells (DC) subsets, mononuclear cells and CD28 co-stimulatory molecule expression. The results showed that: G-PB lymphocytes, CD3, CD4, CD8 T lymphocytes, DC1, DC2, as well as the content of mononuclear cells, CD4/CD8 ratio higher than the G-BM (P lt;0.001). G-PB T-lymphocyte proliferation higher than G-BM (P lt;0.05); each rose G-PB graft T-lymphocytes secrete cytokines IFN-γ, IL-4 of the volume was significantly higher than G - BM, and G-PB in the IL-4/IFN-γ ratio is less than G-BM (P lt;0  001), DC2 and T-lymphocyte ratio is also lower than G-BM (P lt;0.01); while the G - PB, CD4, CD8 cell percentage and CD28 expression were higher than the overall expression of G-BM (P lt;0  001). Conclusion: rhG-CSF in vivo induced by G-PB and G-BM produced by low-reactive T cell immunity there is a difference, but the difference between G-PB and G-BM transplantation graft-versus-host disease (GVHD) incidence and immunological basis of varying degrees.
