Abstract
Background:Thisstudyexploredco-receptorusageandpredictionofV3genotypingalgorithmsinHIV-1subtype
B’frompaidblooddonorsexperiencedanti-retroviraltherapyinChinesecentralprovinceinordertodesign
effectivelytherapeuticregimen.
Methods:HIV-1strainswereisolatedintreatmentHIV-1infectionsandtreatment-na?veHIV-1infections,thenco-
receptorusageofHIV-1strainswasidentifiedbasedonGhostcelllinesusingflowcytometry.HIV-1V3regionwas
amplifiedandsubmittedintoweb-server(WebPSSMandgeno2pheno)topredictHIV-1co-receptorusage.The
feasibilityofpredictionHIV-1usagewithWeb-serverassaywasanalyzedbycomparingpredictionofV3genotyping
algorithmswithHIVphenotypeassaybasedonGhostcellline.
Results:45HIV-1strainsand114HIV-1strainswereisolatedfromHIV-1infectionsexposedanti-retroviraltherapy
andtreatment-na?ve,respectively.41%clinicalvirusesfromARTpatientsand18%fromtreatment-na?vepatients
usedCXCR4asco-receptor.ThenetchargeintheV3loopwassignificantlydifferenceinbothgroups.The
sensitivityandspecificityforpredictingco-receptorcapacityis54.6%and90.0%on11/25rule,50.0%and90%on
Web-PSSMx4r5,68.2%and40.0%onGeno2pheno[co-receptor]
.
Conclusion:Dual/mixed/X4co-receptorutilizationwashigherinARTpatientsthantreatment-na?vepatients.Itis
shouldpaidattentiontopredictingHIV-1co-receptorusagebasedonV3genotypingalgorithmsinHIV-1subtype
B’frompaidblooddonorsexperiencedanti-retroviraltherapyinChinesecentralprovince.
