Genetic alterations, inflammatory and angiogenicprocesseshavebeenidentifiedthroughouttumorprogression; however,treatmentstillremains palliativefor mostcases.Biologicalresearchonparametersinfluencingcellsurvival,invasionandtumorheterogeneityidentifiedseveral cytokinesinterferinginCNSinflammation,oxidativestressandmalignanttransformation,includingTNF-superfamily(TNFSF) members.Inthisreport weperformed ameta-analysisofpublicgene-array dataontheexpressionofagroupofTNFSF ligands(BAFF,APRIL,TWEAK)andtheirreceptors(BAFF-R,TACI,BCMA,Fn14)ingliomas.Inaddition,weinvestigatedby immunohistochemistry(IHC)thetumorcells’expressionoftheseligandsandreceptorsinaseriesof56gliomasofdifferent grade.WeshowthatinIHC,BAFFandAPRILaswellastheircognatereceptors(BCMA,TACI)andFn14expressioncorrelate withtumorgrade.Thisresultwasnotevidencedinmicro-arraysmeta-analysis.Finally,wedetectedforthefirsttimeFn14, BAFF,BCMAandTACIinglioma-relatedvascularendothelium.Ourdata,combinedwithourpreviousreportingliomacell lines,suggestaroleforthesereceptorsandligandsingliomabiologyandadvancethesemoleculesaspotentialmarkersfor theclassificationofthesetumorstotheproliferative,angiogenicorstem-likemolecularsubtype.
