Here,weconstructaquantitative,predictivetheoryofsolidtumorgrowth,metabolicrate,vascularizationandnecrosisthat integrates the relationships between these properties. To accomplish this, we develop a comprehensive theory that describestheinterfaceandintegrationofthetumorvascularnetworkandresourcesupplywiththecardiovascularsystemof thehost.Ourtheoryenablesaquantitativeunderstandingofhowcells,tissues,andvascularnetworksacttogetheracross multiplescalesbybuildingonrecenttheoreticaladvancesinmodelingbothhealthyvasculatureandthedetailedprocesses ofangiogenesisandtumorgrowth.Thetheoryexplicitlyrelatestumorvascularizationandgrowthtometabolicrate,and yieldsextensivepredictionsfortumorproperties,includinggrowthrates,metabolicrates,degreeofnecrosis,bloodflow ratesandvesselsizes.Besidesthesequantitativepredictions,weexplainhowgrowthratesdependoncapillarydensityand metabolicrate,andwhysimilartumorsgrowslowerandoccurlessfrequentlyinlargeranimals,sheddinglightonPeto’s paradox.Variousimplicationsforpotentialtherapeuticstrategiesandfurtherresearcharediscussed.
