ActionsofaProlineAnalogue,L-Thiazolidine-4- CarboxylicAcid(T4C),onTrypanosomacruzi Anah?′Magdaleno,Il-YoungAhn,LisvaneSilvaPaes,ArielM.Silber* DepartamentodeParasitolog?′a,InstitutodeCie?nciasBiome′dicas,UniversidadedeSa?o Paulo,Sa?o Paulo,Brazil Abstract ItiswellestablishedthatL-prolinehasseveralrolesinthebiologyoftrypanosomatids.InTrypanosomacruzi,theetiological agent of Chagas’ disease, this amino acid is involved in energy metabolism, differentiation processes and resistance to osmoticstress.Inthisstudy,weanalyzedtheeffectsofinterferingwithL-prolinemetabolismontheviabilityandonother aspects of the T. cruzi life cycle using the proline analogue L- thiazolidine-4-carboxylic acid (T4C). The growth of epimastigoteswasevaluatedusingdifferentconcentrationsofT4Cinstandardcultureconditionsandathightemperature oracidic pH. Wealso evaluated possible interactions ofthis analogue withstress conditions suchasthoseproduced by nutrientstarvationandoxidativestress.T4Cshowedadose-responseeffectonepimastigotegrowth(IC50=0.8960.02mM at28uC),andtheinhibitoryeffectofthisanaloguewassynergistic(p,0.05)withtemperature(0.5460.01mMat37uC).T4C significantlydiminishedparasitesurvival(p,0.05)incombinationwithnutrientstarvationandoxidativestressconditions. Pre-incubationoftheparasiteswithL-prolineresultedinaprotectiveeffectagainstoxidativestress,butthiswasnotseenin thepresenceofthedrug.Finally,thetrypomastigoteburstingfrominfectedmammaliancellswasevaluatedandfoundto beinhibitedbyupto56%whencellsweretreatedwithnon-toxicconcentrationsofT4C(between1and10mM).Allthese datatogethersuggestthatT4Ccouldbeaninterestingtherapeuticdrugifcombinedwithothersthataffect,forexample, oxidativestress.Thedataalsosupporttheparticipationofprolinemetabolismintheresistancetooxidativestress.
