PrincipalFindings:WecannowdemonstratethattheactivationofRasrequires,inaddition,theessentialparticipationof ezrin,radixinand/ormoesin(ERM),afamilyofactin-bindingproteins,andofactin.Disruptingeithertheinteractionofthe ERMproteinswithco-receptors,down-regulationofERMproteinsbysiRNA,expressionofdominant-negativemutantsof theERMproteinsordisruptionofF-actin,abolishesgrowthfactor-inducedRasactivation.Ezrin/actincatalyzestheformation ofamultiproteincomplexconsistingofRTK,co-receptor,Grb2,SOSandRas.WealsoidentifybindingsitesforbothRasand SOSonezrin;mutationsofthesebindingsitesdestroytheinteractionsandinhibitRasactivation.Finally,weshowthatthe formationoftheezrin-dependentcomplexisnecessarytoenhancethecatalyticactivityofSOSandtherebyRasactivation. Conclusions: Taking these findings together, we propose that the ERM proteins are novel scaffolds at the level of SOS activitycontrol,whichisrelevantforbothnormalRasfunctionanddysfunctionknowntooccurinseveralhumancancers.
