ActivationoftheSMU.1882TranscriptionbyCovRin Streptococcusmutans PatrickChong.¤,ParthoChattoraj.,IndranilBiswas* DepartmentofMicrobiology,MolecularGeneticsandImmunology,UniversityofKansasMedicalCenter,KansasCity,Kansas,UnitedStatesofAmerica Abstract InStreptococcusmutans,theglobalresponseregulatorCovRplaysanimportantroleinbiofilmformation,stress-tolerance response,andcariesproduction.WehavepreviouslyshownthatCovRactsasatranscriptionalrepressorbybindingtothe upstreampromoterregionsofitstargetgenes.Here,wereportthatinvivo,CovRactivatesthetranscriptionofSMU.1882, whichencodesasmallpeptidecontainingadouble-glycinemotif.WealsoshowthatSMU.1882istranscriptionallylinkedto comA that encodes a putative ABC transporter protein. Several genes from man gene clusters that encode mannose phosphotranferase system flank SMU.1882 -comA genes. Genomic comparison with other streptococci indicates that SMU.1882isuniquelypresentinS.mutans,whilethemanoperonisconservedamongallstreptococci,suggestingthata geneticrearrangementmighthavetakenplaceatthislocus.Withtheuseofatranscriptionalreportersystemandsemi- quantitativeRT-PCR,wedemonstratedthetranscriptionalregulationofSMU.1882byCovR. InvitrogelshiftandDNaseI foot-printinganalyseswithpurifiedCovRsuggestthatCovRbindstoalargeregionsurroundingthe-10regionoftheP1882 . Using this information and comparing with other CovR regulated promoters, we have developed a putative consensus bindingsequenceforCovR.AlthoughCovRbindstoP1882,invitroexperimentsusingpurifiedS.mutansRpoD,E.coliRNA polymerase,andCovRdidnotactivatetranscriptionfromthispromoter.Thus,wespeculatethatinvivo,CovRmayinterfere withthebindingofarepressororrequiresacofactor.
