Thepossibility of ongoingtransmissionbetweenhumansandtheirlivestockhasnotbeensystematicallyexamined. Methodology/Principal Findings: To examine ongoing interspecies transmission, we conducted a prospective cross- sectionalcohortstudyof68familiesandtheirlivestock.Stoolspecimenswerecollectedfrom154peopleand115livestock; GBS was detected in 19 (12.3%) humans and 2 (1.7%) animals (bovine and sheep). Application of multilocus sequence typing(MLST)identified8sequencetypes(STsorclones),withSTs1and23predominating.Therewere11familiesinwhich twomemberssubmittedstoolsandatleastonehadGBScolonization.In3ofthesefamilies,bothmembers(consistingof couples)werecolonized,yieldingaco-colonizationrateof27%(95%CI:7%–61%).Twoofthesecoupleshadstrainswith identicalMLST,capsule(cps)genotype,susceptibility,andRAPDprofiles.Onecoupleco-colonizedwithST-1(cps5)strains alsohadabovinecolonizedwiththeidenticalstraintype.Onmultivariateanalysisofquestionnairedata,cattleexposure wasapredictorofGBScolonization,witheachunitincre
