We hypothesized that T-cell and antibody responses to the vaccines are functions of pre-existing host immunity against influenzaantigens. Methodology/Principal Findings: During the 2004 and 2005 influenza seasons, we have collected data on cellular and humoralimmunereactivitytoinfluenzavirusinbloodsamplescollectedbeforeandafterimmunizationwithinactivatedor liveattenuatedinfluenzavaccinesinhealthychildrenandadults.Wefirstusedcross-validatedlassoregressiononthe2004 datasettoidentifyagroupofcandidatebaselinecorrelateswithT-cellandantibodyresponsestovaccines,definedasfold- increase in influenza-specific T-cells and serum HAI titer after vaccination. The following baseline parameters were + examined:percentagesofinfluenza-reactiveIFN-c cellsinTandNKcellsubsets,percentagesofinfluenza-specificmemory B-cells,HAItiter,age,andtypeofvaccine.Thecandidatebaselinecorrelateswerethentestedwiththeindependent2005 dataset.Baselinepercentageofinfluenza-specificIFN-c CD4T-cellswasidentifiedasasignificantcorrelateofCD4andCD8 + T-cellresponses,withlowerbaseline
