Methodology/PrincipalFindings:WithRT-qPCRandChIPassays,weinvestigatedtranscriptionalregulationoftheS-phase specifichistonegenesandfoundthattheacetylationlevelofhistonesoncorehistonegenepromotersfluctuatedduring cellcycleinapatternsimilartoRNApolymeraseIIassociation.Further,weshowedthatCBP/p300andSIRT1wererecruited tohistonegenepromotersinanNPAT-dependentmanner,knockdownofwhichaffectedhistoneacetylationonhistone genepromotersandhistonegenetranscription. Significance: These observations contribute to further understanding of the mechanism by which the expression of canonicalhistonegenesisregulated,andalsoimplicatealinkbetweenhistoneexpressionandDNAdamagerepairandcell metabolism.
