Methodology/Principal Findings: Usingautomatedbehaviouralmonitoring,acapillaryfeedingassayandpharmacological treatments,weexaminedtheeffectofdietaryyeastandsucroseuponDrosophilasleep-wakebehaviourforthreeconsecutive days.Wefoundthatdietaryyeastdeconsolidatedthesleep-wakebehaviouroffliesbypromotingarousalfromsleepinmalesand shorteningperiodsoflocomotoractivityinfemales.Wealsodemonstratethatarousalfromnocturnalsleepexhibitsasignificant ultradianrhythmicitywithaperiodicityof85minutes.Increasingthedietarysucroseconcentrationfrom5%to35%hadnoeffect ontotalsucroseingestionperdaynoranyaffectonarousal,howeveritdidlengthenthetimethatmalesandfemalesremained active.Higherdietarysucroseledtoreducedtotalsleepbymalebutnotfemaleflies.Locomotoractivitywasreducedbyfeeding fliesMetformin,adrugthatinhibitsoxidativephosphorylation,howeverMetformindidnotaffectanyaspectsofsleep. Conclusions:Weconcludethatarousalfromsleepisunderultradiancontrolandregulatedinasex-dependentmannerby dietaryyeastandthatdietarysucroseregulatesthelengthoftimethatfliessustainperiodsofwakefulness.Thesefindings highlightDrosophilaasanimportantmodelwithwhichtounderstandhowdietimpactsuponsleepandwakefulnessin mammalsandhumans.
